PE-22-28 is a synthetic peptide derived from spadin that selectively blocks the TREK-1 potassium channel — a target heavily implicated in depression, mood regulation and synaptic plasticity. By inhibiting TREK-1, it removes a brake on serotonergic neurons in the dorsal raphe, producing antidepressant-like effects in preclinical work within days rather than the weeks typical of SSRIs. Additional research points to neurogenesis in the hippocampus, BDNF upregulation and cognitive-protective effects after ischemic injury. Of interest as a fast-acting mood-support compound that does not act through monoamine reuptake inhibition. Delivered as a 10mg / 3mL cartridge for precise micro-dosing on a standard insulin pen. Typically run in short evening or AM cycles.
Dosage
Start 0.3mg SubQ daily; after 1 week double to 0.6mg daily (10 clicks starter → 20 clicks on a standard insulin pen — 10mg/3mL cartridge delivers ~0.033mg per click)

Clinical Research
PE 22-28, a shorter analog of spadin, retains the antidepressant properties of spadin by blocking TREK-1 channels, with effects observed after only 4 days of treatment in animal models — a marked acceleration compared with classical antidepressants.

Spadin — the parent peptide of PE-22-28 — was identified as a selective TREK-1 blocker producing antidepressant-like effects faster than fluoxetine in preclinical models. PE-22-28 is the optimised analogue; human clinical data are not yet published.
